Portugal is set to play an important role in an international clinical research programme investigating a new treatment approach for head and neck cancer. The study will evaluate amivantamab, an investigational antibody therapy developed by Johnson & Johnson, in patients with recurrent or metastatic head and neck squamous cell carcinoma, a disease that continues to present significant therapeutic challenges worldwide.
The Phase III trial is expected to enrol approximately 500 participants across multiple countries, with Portugal contributing a cohort of patients through five participating hospitals. The Portuguese centres involved in the study include Hospital de Portimão, Hospital CUF Descobertas, Hospital de Gaia-Espinho, IPO Porto and other selected oncology units, reinforcing the country's growing involvement in international cancer research.
Amivantamab is a bispecific antibody designed to target two biological pathways frequently associated with tumour growth and treatment resistance: epidermal growth factor receptor (EGFR) and mesenchymal-epithelial transition factor (MET). Researchers believe that simultaneously inhibiting these pathways may help slow disease progression while enhancing anti-tumour activity.
Interest in the therapy has increased following encouraging findings from earlier clinical studies. Data presented from the OrigAMI-4 programme demonstrated meaningful clinical activity in patients with advanced head and neck cancers, including tumour shrinkage in a substantial proportion of participants. Investigators also reported durable responses in some patients whose disease had previously progressed despite standard treatments such as chemotherapy and immunotherapy.
One notable aspect of the treatment is its administration method. Unlike many cancer therapies that require prolonged intravenous infusions, amivantamab is delivered through a subcutaneous injection, potentially improving convenience for patients and reducing treatment burden within healthcare settings.
Head and neck squamous cell carcinoma accounts for the majority of head and neck cancer cases globally and is often associated with poor outcomes when the disease recurs or spreads beyond its original site. Despite advances in surgery, radiotherapy, chemotherapy and immunotherapy, many patients continue to face limited treatment options once conventional therapies fail.
Researchers involved in the international programme emphasise that the ongoing study is designed to determine whether the addition of amivantamab to current standards of care can improve outcomes for patients with advanced disease. While further evidence is required before definitive conclusions can be drawn, the launch of the trial in Portugal represents an important step in expanding access to innovative cancer research and exploring new therapeutic possibilities for a challenging form of cancer.
